Proteinase K for Protein Digestion

Proteinase K is a highly active endopeptidase designed for nonspecific protein degradation and nucleic acid purification in the most sensitive applications. With exceptional purity and PCR-grade quality, Proteinase K ensures reliable performance in workflows requiring the removal of proteins and the inactivation of nucleases, making it ideal for preparing high-quality PCR templates.

This robust enzyme is stable over a broad pH range (4–12.5) and retains full activity for several hours at pH 6.5–9.5. Its activity is further enhanced by denaturing agents like SDS and urea, with SDS increasing activity up to seven-fold. Proteinase K is available as a stable lyophilized powder or an aqueous solution, offering flexibility for various experimental needs.

Quality

Protein Concentration
The protein concentration of soluble Proteinase K is analyzed by Biuret assay (20 mg, +/- 2 mg).

Protease Activity
The activity is analyzed with hemoglobin as substrate at 37°C (>600 U/ml).

Absence of nuclease activity
The enzyme is tested for absence of endonuclease and nicking activities.

RNase contamination
The enzyme is tested for absence of RNase activity.

DNA contamination
The enzyme is tested for DNA (threshold 10 pg/mg).

Bioburden
The enzyme is tested for biodurden (<1 CFU/ml).
